L-theanine has become a go-to supplement for people looking for a gentler alternative to sleep aids or anti-anxiety medication. It’s naturally found in tea, widely available as a standalone supplement, and marketed as calming without being sedating. But how much of this reputation holds up under actual research? Here’s what current studies show.
What is L-theanine?
L-theanine is a naturally occurring amino acid found primarily in the tea plant Camellia sinensis, as well as in some mushrooms. Its long history in tea consumption, combined with traditional beliefs about its calming effects, has driven substantial scientific interest in how it affects the brain and nervous system (Kilpatrick, 2026).
What the evidence shows for anxiety and stress
The research on L-theanine and stress is fairly consistent. A 2026 mini-review published in Frontiers in Nutrition, which summarized findings from systematic reviews and meta-analyses of human randomized controlled trials, concluded that L-theanine reduces both self-reported and physiological markers of stress and anxiety under stress-inducing conditions, at doses of 200 to 400 mg per day, without causing sedation (Kilpatrick, 2026).
Individual trials support this pattern:
- A randomized, placebo-controlled, crossover trial in 30 healthy adults found that four weeks of L-theanine (200 mg/day) significantly reduced scores on the State-Trait Anxiety Inventory, a standard anxiety measurement tool, along with depression-related symptoms, compared to baseline (Hidese et al., 2019).
- A study using an acute cognitive stress task found that 200 mg of L-theanine significantly reduced participants’ subjective stress and anxiety response compared to placebo. This was accompanied by measurable reductions in physiological stress markers, including heart rate and secretory immunoglobulin A, suggesting L-theanine may reduce the body’s sympathetic nervous system response to acute stress, not just the subjective feeling of it (as cited in Anti-Stress, Behavioural and Magnetoencephalography Effects study, n.d.).
- A separate randomized, triple-blind, placebo-controlled study found that a single dose of L-theanine significantly increased frontal alpha brainwave activity three hours after dosing during an EEG recording. Increases in alpha power are generally associated with a relaxed mental state (PMC, 2021).
It’s worth noting that not every study finds a clear effect. One trial using a more intense stressor (electric shock) found no significant anti-anxiety benefit from either L-theanine or a comparison anti-anxiety medication, suggesting L-theanine’s calming effect may be relatively mild and more effective against everyday stress than against very intense anxiety triggers (as cited in Anti-Stress, Behavioural and Magnetoencephalography Effects study, n.d.).
What the evidence shows for sleep
The most comprehensive evidence comes from a 2025 systematic review that analyzed 13 eligible trials involving 550 participants, examining L-theanine (50 to 900 mg/day) as a standalone intervention for sleep. The review concluded that supplementation with 200 to 450 mg per day appears to be a safe and effective way to support healthy sleep in adults, with reported benefits across both objective measurements and participant-reported outcomes, including sleep latency (how long it takes to fall asleep), sleep maintenance, sleep efficiency, and how refreshed people felt upon waking (PubMed, 2025).
Other individual studies back this up:
- The same 30-person, four-week trial noted above also found significant improvements in Pittsburgh Sleep Quality Index (PSQI) scores, a widely used clinical sleep questionnaire, along with specific improvements in sleep latency, sleep disturbance, and reduced use of sleep medication compared to placebo (Hidese et al., 2019).
- A double-blind, randomized, placebo-controlled crossover trial combining L-theanine with a milk-derived ingredient (alpha-S1-casein tryptic hydrolysate) also used the PSQI to assess poor sleep quality, reflecting how frequently this tool is used as a standard measure in L-theanine sleep research (PMC, 2022).
Importantly, unlike many sleep aids, L-theanine at these doses has not been shown to cause sedation. This is considered one of its more distinctive features: it appears to promote a calmer, more relaxed state that supports natural sleep onset, rather than directly sedating the user (Kilpatrick, 2026).
How L-theanine may work
While the exact mechanism is still being studied, L-theanine’s effects on alpha brainwave activity, a pattern associated with wakeful relaxation and states like meditation, are one of the more consistently observed physiological findings (PMC, 2021). Researchers have also documented reductions in physiological stress indicators like heart rate and immune stress markers following L-theanine intake, suggesting it may act on the body’s stress response system rather than working purely as a psychological placebo effect (Anti-Stress, Behavioural and Magnetoencephalography Effects study, n.d.).
Effective dosage
Across the research reviewed, most studies used daily doses in a fairly consistent range:
- For anxiety and stress: 200 to 400 mg per day (Kilpatrick, 2026)
- For sleep support: 200 to 450 mg per day, based on the most comprehensive systematic review to date (PubMed, 2025)
Trials have tested doses as low as 50 mg and as high as 900 mg per day, but the strongest and most consistent evidence clusters around the 200 to 400 mg range.
Safety profile
L-theanine has a favorable safety record in the research reviewed. Pre-clinical studies indicate it is neither mutagenic nor carcinogenic and has low toxicity, with a median lethal dose far beyond typical supplemental amounts (PMC, 2022). Human trials have generally reported it as well tolerated, with safety monitoring in some studies including vital signs, blood chemistry, and blood counts showing no significant adverse events tied to supplementation (as cited in Examining the effect of L-theanine on sleep, 2025).
Limitations worth knowing
The evidence base, while generally positive, isn’t without gaps:
- Many trials have relatively small sample sizes, often around 30 participants, which limits how confidently findings can be generalized.
- Some included studies in systematic reviews were open-label rather than placebo-controlled, a design that carries a higher risk of bias, though researchers have noted their results were broadly consistent with the more rigorously designed, double-blind, placebo-controlled trials (Examining the effect of L-theanine on sleep, 2025).
- Most existing research focuses on generally healthy adults rather than people with diagnosed anxiety disorders or clinical insomnia, so it’s less clear how well these findings extend to more severe or chronic cases.
- Researchers have specifically called for more long-term, double-blind, placebo-controlled trials to confirm findings, particularly for stress and sleep outcomes measured over more than a few weeks (Examining the effect of L-theanine on sleep, 2025).
Bottom line
Among natural sleep and anxiety supplements, L-theanine has a comparatively solid base of human research behind it. Multiple randomized controlled trials and systematic reviews support real, measurable reductions in stress and anxiety symptoms, along with improvements in sleep quality and sleep onset, generally at doses between 200 and 450 mg per day, without the sedating effects associated with many sleep medications. That said, most studies involve small groups of generally healthy adults over relatively short periods, so it’s reasonable to view L-theanine as a promising, low-risk option worth discussing with a healthcare provider, rather than a guaranteed fix for clinical anxiety or diagnosed insomnia.
